Consta no site Museu de Vila Maria que a data oficial de fundação da Vila Medeiros é 11/10/1912, pois as terras adquiridas por José de Medeiros foram nessa data registradas pelo escrivão de paz e tabelionato de notas do distrito de Santana. Assim o bairro completou 99 anos.

Às margens da antiga Estrada Real, que ligava a Guarulhos, essas terras foram objeto de plantio de milho, feijão, batata-doce, cana e também uva. Conta a história que em tempos passados, antes de se transformar em um bairro predominantemente residencial, chegou a abrigar grande produção de leite de gado holandês, obra daquele fundador, José de Medeiros.
A criação de gado durou até 1924, quando houve o loteamento em chácaras de 5, 10 e 20 mil m2. Entre os primeiros compradores estavam Juvenal Sayão, Antonio Fernandes e Rafael Purisi. Na "Cantina do Medeiros" eram comercializados queijos, lingüiça e vinho de produção local. O tino comercial e artístico do fundador trouxe para a região uma pequena romaria de turistas, atraídos pelas quadras de bocha e de malha construídas ao lado da cantina. A produção artesanal de violões e bandolins no local contribuiu para o trânsito de visitantes.
Outra curiosidade foi a criação de um "sanitário de lavagem de carrapatos" na região, para a higienização do gado que existia ali. Muitas pessoas levavam seus animais para esse tratamento no local. José Munhoz Bonilha, ao auxiliar o proprietário Medeiros no loteamento da região, ganhou terras em comissão nas proximidades da atual avenida Conceição. Surgia assim o bairro Vila Munhoz.

No final da década de 1940 os primeiros comércios chegaram à av. N. Sra. do Loreto, hoje centro da Vila Medeiros. Até a década de 1950 era comum os moradores caminharem até a estação Tucuruvi, para pegar o trenzinho da Cantareira. Só em 1954 a antiga empresa de ônibus municipal, a CMTC, criou a linha Largo da Concórdia - Vila Medeiros, porém bastante precária em número de carros e de horários. O que mais acudia os moradores eram os caminhões, mais freqüentes. A empresa de ônibus Alto do Pari, a partir de 1956, ampliou a quantidade de linhas para a região.
Hoje duas entidades se destacam na prestação de serviço na região: A ASSORAVIM - Associação Reivindicativa e Assistencial de Vila Medeiros, e o Rotary Club São Paulo Vila Medeiros. A ASSORAVIM foi fundada em 24/04/1966 por um grupo de comerciantes, que se reuniam na "Farmácia Boa Sorte". A entidade realiza trabalhos na área social, voltados à população carente.
O Rotary Vila Medeiros foi fundado em 08/06/2004. Seu presidente Jairo Gomes da Fonseca informa que o clube tem atualmente 19 membros. Todo mês de Abril a entidade realiza um mutirão de saúde e cidadania. No evento deste ano foram feitos 5.040 atendimentos a população carente. O projeto Rumo, de orientação vocacional para os jovens que irão ingressar no mercado de trabalho, atua todo ano em uma escola pública da região. A primeira reunião do mês acontece na sede da ASSORAVIM. As demais, todas as 2ªs-feiras, 20:30h, são realizadas na Churrascaria Super Grill, no pátio do estacionamento do Carrefour Vila Maria. Fonseca cita como ações sociais relevantes o apoio ao Projeto da Amizade, na Vila Constança, e à CEI Sagrada família, no Jardim Brasil, creche que educa 145 crianças.
